In this study, histological and histochemical properties and density oL mast cells were investigated in the skin of dog. For this purpose skin biopsies ./i'om nine dogs with no clinical or histological evidence ol skin diseases were examined. Biopsies of the dogs were obtained from the lateral abdomen. Each specimen was divided into two pieces which were placed in both neutral butlered formalin (NBF) and isotonic formalin acetic acid .fixatives (IFAA). To demonstreate the mast cells, tissue sections were stained with toluidine blue (TB), alcian blue / salranin O (AB/SO) and Berberine sulphate staining methods. The mast cells were observed in subepidermal region, around the blood vessels, sweat glands, sebaceous glands and hair follicules. In IFAA fixed tissue, the mast cells could not be demonstreated sufficiently with AB/SO staining method. The mast cells were stained as AB( +), AB/SO( +) andfrequently SOr +) in NBFfixed tissues. The hepw'in containing mast cells were also demonstreated with Berberine sulphat staining methods in subepidermal region, around the blood vessels, sweat glands, sebaceous glands and hair follicules. With TB staining method, the mast cell density were detected as 27.80 :t 1.68 mast cells /mm2 in NBF fixed tissues and 29.09 :t 3.16 mast cells /mm2 in IFM .fixed tissues. No statistically signijicant diflerences were found between the number of TB (+) mast cells in the NBF and IFAAjixed tissues.
