_??_ ypothesis was advanced that UDP-galactose would donate galactosyl residues to sucrose to f orm raffinose at first and then followed by higher members of this series. This report is the outcome of the experimental verification of this hypothesis and describes the bio synthesis of an oligosaccharide, chromatogra phically identical with raffinose, from UDPgalactose and sucrose by an ezyme preparation from immature soybeans. UDP-galactose was prepared essentially by the method of Wiesmeyer and Jordan4) by using Escherichia coli, strain C 7 SM, which was kindly furnished by Prof. Ushiba of Keio University, and purified by chromatography on a column of Dowex 1 .x 4 (Cl-, 200-400 mesh) by the stepwise elution system of NaCl in 0.01 N HCl,5) followed by rechromatography on a similar but smaller column by a linear gradient elution system of increasing con centrations of NaCl in 0.01N HCl.
