Aim: To compare the efficacy and safety of a fixed-dose combination of diclofenac 50 mg and paracetamol 500 mg versus paracetamol 500 mg alone in the management of dental pain following tooth extraction. Materials and Methods: A randomized, open-label study of diclofenac 50 mg + paracetamol 500 tablets (Group A) versus paracetamol 500 mg tablets (Group B) was conducted in 50 patients after extraction of tooth with moderate or severe pain with enrollment duration of 4 days per patient. Adverse drug reaction (ADR) in each group was also analyzed by Naranjo’s scale and statistically analyzed. Results: Pain intensity decreased significantly at all the time points in both the groups (P < 0.01). Significantly higher differences in pain intensity were achieved with Group A compared to Group B at 2, 4, 6, 8 and 24 h after extraction (P < 0.05). Degree of pain relief for Group A was comparable to that of Group B at different time points (P > 0.05). No rescue medication was required in Group A while in Group B, nine patients required rescue medication. ADR was significantly found in Group A. The physician and patient global assessment scores were more inclined towards the combination treatment. Conclusion: It can be concluded that diclofenac 50 mg + paracetamol 500 mg and paracetamol 500 mg alone are found to be effective in the management of dental pain following tooth extraction with need of rescue medication in Group B. However, combination of diclofenac + paracetamol was found with possible ADR as compared to Group B.
