Within the blood stream, thyroxine is distributed among the plasma proteins according to their relative binding affinities for this material. Thus, circulating thyroxine is principally associated with a moiety intermediate in electrophoretic mobility at pH 8.6 between the a, and a2 globulins (1-6). This "thyroxine-binding protein" of plasma (TBP) 5 has been localized in subfractions of plasma prepared by Method 6 of Cohn, and it has been shown that all subgroups of Fraction IV-4 (especially IV-6 and IV-9) contain TBP greatly in excess of the normal serum proportions (6). In the present studies, the presence of TBP has been demonstrated in extravascular fluids, and Cohn Fractions (7-9) and tissue slices have been employed for in vitro models designed to assess the possible significance of TBP in the partition of thyroxine between cellular and extracellular phases.
