Haploid strains of S. cerevisiae were exposed to ultraviolet irradiation, and mutants were isolated by a modified layer plate procedure (Lederberg and Tatum, 1946), using a complete agar as the recovery layer. Six mutants were found which required adenine, two which required uracil. A seventh adenineless mutant was obtained through the courtesy of S. E. Reaume (Reaume and Tatum, 1949). Genetic analyses were conducted as previously described (Pomper and Burkholder, 1949; Pomper, 1951). Growth conditions were those of Schultz and Atkin (1947), volume and inoculum reduced 50 per cent, using the synthetic medium of Burkholder and Giles (1947). Growth is expressed as per cent absorption in a "lumetron" colorimeter, using a wire-glass filter.
