Pseudomonas aeruginosa and the phytopathogenP. syringae produce the exopolysaccharide alginate, which is a copolymer ofd -mannuronic andl -guluronic acids. One of the key regulatory genes controlling alginate biosynthesis inP. aeruginosa isalgT , which encodes the alternate sigma factor, ς22 . In the present study, thealgT gene product fromP. syringae pv. syringae showed 90% amino acid identity with itsP. aeruginosa counterpart, and sequence analysis of the region flankingalgT inP. syringae revealed the presence ofnadB ,mucA , andmucB in an arrangement virtually identical to that ofP. aeruginosa . AnalgT mutant ofP. syringae was defective in alginate production but could be complemented with wild-typealgT fromP. syringae orP. aeruginosa when expressed intrans . ThealgT mutant also displayed increased sensitivity to heat, paraquat, and hydrogen peroxide (H2 O2 ); the latter two compounds are known to generate reactive oxygen intermediates. Signals for activation ofalgT gene expression inP. syringae were investigated with analgT ::uidA transcriptional fusion. Like that inP. aeruginosa ,algT transcription inP. syringae was activated by heat shock. However,algT expression inP. syringae was also stimulated by osmotic stress and by exposure to paraquat, H2 O2 , and copper sulfate. The latter two compounds are frequently encountered during colonization of plant tissue and may be unique signals foralgT activation inP. syringae .
