SUMMARY: A bioassay was developed for the study of the biochemical induction of homo-karyotic fruiting in Schizophyllum commune. A fruiting-inducing substance (FIS) was incorporated into a small cylindrical agar block which was used to induce the test colony grown on cellophane membrane. The location, time of fruiting and number of fruiting bodies were predictable. This assay system was employed to study the process of fruiting induction. Although colonies up to 9 days old could be induced to fruit, the induced response was restricted in these colonies to hyphae less than 24 h old at the time of FIS exposure. Light was necessary during a period following FIS application for fruiting to occur. High temperature (30°C) was found to inhibit FIS-induced fruiting but did not prevent FIS uptake by the colony.
