Results: PLZ inhibited 97.7 and 99.6% of the Enterobacteriaceae isolates at ≤2 and ≤4 μg/mL, respectively, and PLZ (MIC50/90, 0.5/1 μg/mL) was at least two-fold more active than other aminoglycosides against these isolates (Table). Against CRE (n=50), PLZ (MIC50/90, 0.25/1 μg/mL) displayed more potent activity compared to amikacin (AMK; MIC50/90, 8/32 μg/mL), gentamicin (GEN; MIC50/90, 2/>8 μg/mL) or tobramycin (TOB; MIC50/90, >8/>8 μg/mL). All CRE isolates were inhibited by PLZ at ≤2 μg/mL. PLZ was very active against E. coli (n=657) and K. pneumoniae (n=722), inhibiting 100.0 and 99.9% of these isolates at ≤2 μg/mL. All K. oxytoca (n=177), E. cloacae (52) and Citrobacter spp. (147) isolates were inhibited by PLZ at ≤1 μg/mL. PLZ inhibited 81.1 and 92.5% of the P. aeruginosa isolates at ≤4 and ≤8 μg/mL. PLZ (MIC50/90, 4/8 μg/mL) activity against P. aeruginosa was similar to AMK (MIC50/90, 4/8 μg/mL), but less postent than GEN (MIC50/90, 2/4 μg/mL) and TOB (MIC50/90, 0.5/1 μg/mL). PLZ was very active against coagulase-negative staphylococci and S. aureus (including 15 methicillin-resistant isolates), inhibiting all isolates at ≤0.5 and ≤2 μg/mL, respectively. The activity of PLZ was limited against Acinetobacter spp. (n=45; MIC50/90, 2/16 μg/mL), S. pneumoniae (35; MIC50/90, 32/64 μg/mL) and Enterococcus spp. (32; MIC50/90, 8/64 μg/mL).
