Here we present the nucleotide sequence of the chloroplastACKNOWLEDGEMENTS encoded psbA and trnH genes and intergenic region from cotton Gossypium hirsutum cv. 108-f. psbA codes for 32 kD herbicideW e w o u l d l i k e t 0 t h a n k P r o f D r R-G.Herrmann (FRG) for binding component of thylakoid membrane (also known as QB Providing the probe from spinach. or Dl protein) and trnH codes for tRNA (GUG). It was shown (1) that these genes are cotranscribed as a single operon REFERENCES with post-transcriptional processing. They are localized on 8.4 kb BamHI fragment of cpDNA and wer/cloned as 4.9 and 3.5 \ kb BamHI-PstI fragments using internal PstI site in the psbA gene because of unclonable feature of the whole BamHI fragment. Subsequent attempts to reconstruct complete psbA gene in pUC plasmids failed supporting the hypothesis about the toxicity of its product to bacterial cells.
