During B cell maturation the immunoglobulin loci undergo DNA rearrangements leading to the joining of V and (D)J gene segments (1). The DNA between Vv tained in the genome and JK gene segments is sometimes rea heaa to head fusion product of the V, and JR flanks, the so-called f fragments (e.g. 2,3). Their chromosomal location is relevant to the question of the mechanism of Vj.-JK rearrangement. We have recently reported the structure of an f fragment of the Burkitt lymphoma derived cell line JI (3), which is not the reciprocal product of the V-J joints found in this cell line; it stems from an earlier V-J joining reaction. The chromosomal origin of this f fragment could not be determined unambiguously with Southern blot hybridizations using DNA of somatic cell hybrids because the karyotype of the hybrids is not fully known (3). In the cell line JI the normal chromosome 2 and the translocation chromosomes 2pand 8q+ can be easily distinguished by in situ hybridization(4). The DNA probe m2132-2/2 (probe b in Fig.l of ref.5) which is derived from the 5' flank of J..1 was used in the studies. A positive hybridization signal was detected only on the normal chromosome 2 at the band 2pl2,the site of the Kappa locus (Fig.l).
