A liquid chromatographic method is described for the determination of 9 amines in fish: ammonia, methylamine, tryptamine, putrescine, cadaverine, histamine, tyramine, spermidine, and spermine. The amines are extracted with 0.2M perchloric acid and labeled with dansyl chloride. After labeling, proline is added to neutralize excess dansyl chloride. The amines are separated on a Kromasil C18 column using a water-acetonitrile elution gradient. Handling time is restricted to 40 min, and an injection is possible every 30 min, thus allowing automation and routine analysis.
