Dear Sir: In our work (1) we have made use o f Stein and Lieb's (2) approach to obtain values of ratios o f molecular rate constants related to t ranspor t steps. In o rd e r to rationalize ou r results we assumed at the t ime that in equi l ibr ium-exchange conditions the concent ra t ion o f the carrier-substrate complex was h igher at the external surface of the cell and that the tu rnover rate o f influx was propor t ionally smaller than that o f eff lux. This implied a d i f fe ren t resistance o f the membrane to the inward and outward movements , respectively, o f the loaded carrier. T h e r e is, however , ano the r possibility which we have not previously considered, namely, that the loaded carriers are equally distr ibuted across the membrane but that the un loaded carriers are not. T h a t this is in fact the case for the ur idine t ranspor t system of h u m a n red blood cells we shall a t tempt to demonst ra te here . Briefly, f rom Stein and Lieb's work (2) it can be shown that at steady state the concentrat ion o f the car r ier at the inner side of the m e m b r a n e is given by
