One of the main disadvantages of cements based on Ca(OH) 2 /iodoform is the potential risk of intracanal absorption. Polydimethylsiloxane-based cement have been shown to be less cytotoxic and have better intracanal flow and viscosity as well as being less cytotoxic than epoxy resin-based cement. Titanium diisopropoxide bis (acetylacetonate) (Tyzor AA) is a catalyst that due to its properties could improve the handling of polydemethylsiloxane without affecting its sealing properties. In the present study, we evaluated the cytotoxicity and biocompatibility of a new hydrophobic endodontic sealant based on polydimethylsiloxane-Tyzor AA as a catalyst. A study was carried out in human fibroblasts performing an MTT assay to evaluate cell viability and an  in vivo  rat model for its biocompatibility at 15 and 30 days. The data and the histological evaluation were analyzed using a Cochran Q test with a value of  p  &lt; 0.05. Two freshly mixed formulations showed (86.82%) and formulation V4 (24 h) set showed cell viability (86.98%). The  in vivo  test of formulation V1 (30 days) showed a value of 0.416 in Cochran's  Q  test. Therefore, it is concluded that the new sealant showed acceptable biocompatibility according to international standards.
