Abstract A new class of tandem repetitive DNA element was identied from Triticeae genome. A genomic DNA library was constructed from Aegilops tauschii by cleavage with KpnI. The novel repetitive sequence pAt2, which was enriched in the D genome, was successfully identied. Southern blot hybridization showed that pAt2 occurs as a tandem array. Analysis of pAt2 showed specicity to certain Triticeae genomes. The repetitive clone pAt2 was localized on subtelomeric regions of seven rye chromosomes. The sequence of pAt2 did not share any identity with known repetitive elements, especially the representative subtelomeric sequence 350-family, or any known class I or class II transposable elements. Only several DNA segments in BACs of mice, human, and zebrash showed partial similarity to the repetitive DNA element.
