Although the development of DNA sensors, including gold-based electrodes, has made rapid progress, it is essential to characterize DNA-gold surfaces to better understand their operation and improve performance. Recently, a novel polymer gold electrode (PGE) that shows promising performance as DNA biosensor has been designed. Herein, superior electrochemical characteristics of PGE were further studied and reported. For example, a striking difference of potential between Ru(NH 3 ) 63+  adsorbed onto DNA and in solution was observed, indicating the presence of distinct electron transport pathways of the two states of Ru(NH 3 ) 63+ . Because of the superior electrochemical properties of the PGE, discrimination of single nucleotide mismatch was achieved without signal amplification or sensitive labeling.
