Significance MYC is a transcriptional regulator that controls much of the coding and noncoding transcriptome. It is also an oncoprotein that functions as a driver in numerous human cancers. The mechanism of this oncogenic activity is not known, but it probably involves specific MYC-regulated target genes. Here we systematically identify and characterize MYC-regulated, long noncoding RNAs (lncRNAs) that are required for MYC-driven cellular proliferation in human lymphoid cells. We use targeted transcriptional repression, adding new tools to this technology that will facilitate the functional analysis of any transcriptional regulator.
