Significance The process by which cells differentiate is central to multicellular development and cancer. Dramatic gene expression changes mediate this complex process, which involves the termination of proliferation and the acquisition of distinct cell-specific features. We identified a transcription factor, MYB DOMAIN PROTEIN 36 (MYB36), that regulates this developmental transition in theArabidopsis thaliana root endodermis. Differentiated endodermis forms a protective waxy barrier called the Casparian strip. We found that MYB36 activates genes involved in Casparian strip formation and represses genes involved in proliferation. Our results suggest that MYB36 is a critical regulator of developmental timing in the root endodermis.
