Research in wound healing is important in medical as well as in social aspects due to the enormous increase in leg ulcers and their costs in all industrial countries. To continuously study the early regeneration of human epidermis we have established a skin organ culture model. Skin samples were specially (6 mm diameter) trimmed, the central epidermis (3 mm diameter) was removed and the samples were cultured under defined conditions. Early reepithelialization can be studied for ten days. Most samples have healed spontaneously after seven days of culture. Application of allogenic freshly isolated outer root sheath keratinocytes accelerated reepithelialization and differentiation as well. It was demonstrable that the allogenic keratinocytes are integrated into the neoepithelium including its basal layer. Even more these cells undergo mitoses and start to form proper cell-cell-junction within a few hours. Thus a human model has become available to study reepithelialization under a variety of influences and pharmaceutical substances.
