The regression of platelet malondialdehyde (MDA) formation in normal subjects was investigated after a single oral intake of various doses of aspirin (ASA). To increase the sensitivity of MDA determination by the use of thiobar- bituric acid reaction, platelets were incubated with arachi- donic acid. In the large dose group, the regression curve plotted as percent recovery had a initial lag phase (ILP) for 2 days and became linear thereafter. However, in the small dose group it was almost linear without ILP. The difference of the recovery time between these 2 groups was about 2 days. When the MDA formation was not completely inhibited, the degree of the maximal inhibition in the circulating platelets djd not affect the recovery time. Undetectable serum ASA 4 hrs after the intake suggested the effect of residual ASA on the recovery time could be negligible. These data support the concept that megakaryocytes could be affected m vivo by large doses of ASA, but slightly or not by its small doses which affect circulating platelets.
