The reaction of the rat aortic intima to hypertension varies in experimental models having different pathogenetic mechanisms. Thus, thickness, permeability to horseradish peroxidase, mitotic index and density of endothelial cells in the aorta are more marked during hypertensive situations associated with high levels of circulating exogenous or endogenous mineralocorticoids than in situations associated with low levels of these hormones. We have correlated mitotic index and cell density of endothelial cells with the internal diameter of the aorta in early and late phases of 3 models of experimental hypertension: 1) aortic ligature between renal arteries; 2) uninephrectony and Na-rich diet; and 3) uninephrectomy, 0.9% NaCl as drinking fluid and administration of desoxycorticosterone acetate (DOCA). Mitotic index and/or cell density were increased compared to controls: l) early after aortic ligature, and 2) early and late after DOCA. The internal diameter of the aorta, measured morphometrically after perfusion with glutaralde- hyde at the mean pressure of each rat, was increased only late after aortic ligature when mitotic index and cell density were normal and late after DOCA when mitotic index was normal and cell density was high compared to controls. These results show that there is no correlation between the internal surface of the aorta and the variations of mitotic index and/or cell density of endothelial cells during different types of hypertension and suggest that during certain types of hypertension mechanisms other than changes in the aortic endothelial surface stimulate the replication of endothelial cells.
