The accurate estimation of the a-keto acids present in plant tissues is imnportant because these acids occupy a central position in the metabolism of the plant and thus may be vital to our understanding of the physiological conditions which govern the storage ofmany fruits and vegetables. The main uncertainty in the estimation is the stability of the a-keto acids and related compounds in contact with the tissue during the procedure used for killing. Preliminary results on strawberry leaves showed that different amounts of oc-keto acids were obtained, according to the procedure. This observation focused attention on the peculiarly difficult nature ofthe estimation of substances, such as m-keto acids, which are intermediates in the metabolic pathways linking fats, carbohydrates and amino acids. Only small amounts are present under normal physiological conditions and they are probably closely associated with multi-enzyme systems, which are confined for the most part in small subcellular units such as mitochondria and microsomes.
