Optical tomographic imaging of biological specimen bases its reliability onthe combination of both accurate experimental measures and advancedcomputational techniques. In general, due to high scattering and absorption inmost of the tissues, multi view geometries are required to reduce diffuse haloand blurring in the reconstructions. Scanning processes are used to acquire thedata but they inevitably introduces perturbation, negating the assumption ofaligned measures. Here we propose an innovative, registration free, imagingprotocol implemented to image a human tumor spheroid at mesoscopic regime. Thetechnique relies on the calculation of autocorrelation sinogram and objectautocorrelation, finalizing the tomographic reconstruction via a threedimensional Gerchberg Saxton algorithm that retrieves the missing phaseinformation. Our method is conceptually simple and focuses on single imageacquisition, regardless of the specimen position in the camera plane. Wedemonstrate increased deep resolution abilities, not achievable with thecurrent approaches, rendering the data alignment process obsolete.
