Systemin is a tomato peptide hormone that\udpromotes plant defense against plant-chewing pests. This\udoctadecapeptide is released from the C-terminal region of\udprosystemin, a 200 amino acid-long cytosolic precursor.\udHomologues of the tomato prosystemin are present in other\udSolanaceae but not in tobacco. This species does not respond\udto the exogenous application of the tomato systemin. Previously,\udit was shown that the expression of the full-length\udtomato prosystemin cDNA in tobacco affects the proteomic\udrepertoire and increases plant tolerance against phytopathogenic\udfungi. In this work, to evaluate the relevance of\udthe systemin sequence, we generated transgenic tobacco\udplants that express a mutated prosystemin lacking the systemin\udregion. By using proteomics and gene expression\udanalyses, we show that the constitutive expression of the\udtruncated prosystemin altered the proteomic profile of\udtobacco leaves and increased plant resistance against B.\udcinerea. The overlap of the modifications caused by the\udexpression of the full-length and the truncated prosystemin\udindicated that these alterations occur also in the absence of\udthe systemin sequence. Although the cellular mechanisms of\udprosystemin cleavage are elusive, our work demonstrates\udthat the expression of a heterologous cytosolic peptidehormone\udprecursor, irrespective of the presence of its peptide\uddomain, associated with unpredicted changes at the\udproteomic and transcriptional level

The expression of the tomato prosystemin in tobacco induces alterations irrespective of its functional domain
