Cells ofCoscinodiscus granii of about 200 µm diameter were cultivated at 13° to 17° C in 10-l flasks filled with nutrient solution of about 32 ‰ salinity according to v.Stosch &Drebes (1964). Illumination was provided by fluorescent lamps (12 hours per day) in combination with daylight. Brightness and quality of irradiation were subjected to considerable changes (2000–6000 lux), simulating ecological conditions. At a diatom concentration of about 10 cells/ml, the material was sifted off by plankton cloth (meshwidth 50 µm) and rinsed by isotonic LiCl-solution. Further treatment for cell sap preparation was performed as described byKesseler (1967). As inC. wailesii, potassium was accumulated up to about 50 times of its medium concentration, while sodium was reduced to about 10%. Chloride distribution was equal in cell sap and medium. Similar values were obtained inC. wailesii. In addition to the methods employed in this species, nitrite, nitrate and silica were also determined. While NO2′ was detected only in traces, NO3′ was present in considerable amounts (about 0.0225 Val/l). The Estimations of silica, however, were low, seemingly due to the fact, that only orthosilicate and straight chain polymers of not more than 3 Si atoms will react with molybdate. The ready formation of Si-compounds, nonreactive under the conditions of the method employed may help to explain the deviation of the cation: anion relation from stoichiometric unity. The density of artificial sap prepared according to the analytical results exceeds the density of the medium by about 1.2 mg/cm3. It provides, therefore, no satisfactory explanation for the flotation of diatoms.Zusammenfassung1. In Anbetracht der bisher vorliegenden widersprüchlichen Ergebnisse über die chemische Zusammensetzung des Zellsaftes mariner Diatomeen wurden umfangreiche Untersuchungen anCoscinodiscus granii durchgeführt.2. Das Untersuchungsmaterial wurde unter Bedingungen kultiviert, die den Verhältnissen der freien Natur in gewisser Weise entsprachen. Für die Analysen wurde nur frisches Material verwandt, das sich in der logarithmischen Wachstumsphase befand.3. Die Analysenergebnisse zeigen, daß auch beiC. granii K. und Cl′ als intrazelluläre Ionen dominieren. Der Na.-Gehalt beträgt nur etwa 10 % des K.-Gehaltes.4. Die Resultate einer chemischen Untersuchung des Zellsaftes vonC. wailesii werden dadurch im wesentlichen bestätigt; lediglich der damals bestimmte unwahrscheinlich hohe SO4′'-ehalt erwies sich als Irrtum.5. Wider Erwarten ergaben die Silikatbestimmungen nur sehr niedrige Werte. Hierbei ist jedoch zu berücksichtigen, daß mit der üblichen Si-Nachweisreaktion nur Orthokieselsäure und Polymerketten mit maximal 3 Si-Atomen erfaßt werden. Ein nicht unbeträchtlicher Teil des tatsächlich vorhandenen Silikats mag sich so dem Nachweis entzogen haben.6. Die aräometrisch ermittelte Dichte künstlichen Zellsaftes, der nach den Analysenergebnissen hergestellt wurde, betrug 1,0248 g/cm3. Das Kulturmedium hatte demgegenüber nur ein spezifisches Gewicht von 1,0236 g/cm3. Die chemische Zusammensetzung des Zellsaftes allein vermag daher nicht zu erklären, weshalb Diatomeen dennoch fast ausschließlich in oberflächennahen Wasserschichten schwebend anzutreffen sind.
