An F′ factor, FS400, carrying thehis operon, thegnd gene, and therfb gene cluster ofSalmonella typhimurium was isolated. FS400 was introduced into anEscherichia coli strain having a lengthy deletion of thehis gene region. From this strain, Hfr derivatives were isolated which had the F′ factor integrated in thetonB locus near the attachment site of φ80. One of the Hfr strains was lysogenized with a heat-inducible,h mutant of φ80, and from this strain a high-frequency transducing phage carrying thehis genes and thegnd gene ofSalmonella was isolated.
