Despite the principal role of melanosomes is clear, the molecular structure of melanins, the mechanism by which the pigment is produced, and how the pigment is organized within the melanosome remains to be fully understood. A procedure for the analysis of melanin-pigmented skin samples has been set-up. The method is based on alkaline hydrogen peroxide degradation coupled with HPLC and the ultraviolet determination of PTCA for eumelanin and TTCA for pheomelanin. The advantages connected to the degradation conditions and sample handlings were coupled to an improved chromatographic methodology for the simultaneous determination of PTCA and TTCA as representative markers of either eumelanin or pheomelanin.\udA 3D plot representation that combines the analytical parameters (i.e. TTCA and PTCA) in connection to the specific skin phototype is proposed.
