Bone marrow is incubated in colcemid solution (0.1 μg./ml.), treated in hypotonic KCl (.075 M) and fixed in Carnoy's fluid. Chromosomes are spread by a ‘flaming’ technique. Yield of mitotic figures is high and staining quality good.
