Preparation and Properties of an Immunosorbent Column Specific for the Myelin Basic Protein

An immunosorbent column specific for the myelin basic protein (BP) was prepared by coupling purified anti‐BP antibodies to cyanogen bromide (BrCN)‐activated Sepharose 4B. The BP‐immunosorbent column bound BP between pH 4.5 and pH 6.8. In its working range the column bound approximately 400‐475 μg of BP at pH 6.8 and 250 μg at pH 4.5 with recoveries of 72‐77%. The BP‐immunosorbent column could effectively separate BP from simple mixtures of BP and proteins of similar size and charge and from acid extracts of bovine brain. The results indicate that the BP‐immunosorbent column can be used to isolate BP from a mixture of proteins and may be adapted for use in the small‐scale purification of the myelin basic proteins involving a minimum number of steps.
