Methods Human primary pulmonary endothelial cells (HPAEC) and whole pulmonary artery organoid cultures were isolated from human lungs after pneumonectomy and cultivated on gelatincoated flasks. HPAEC were identified by electron microscopy and immunocytological staining with mAb against human v.Willebrand factor. Both HPAEC and organoid cultures were challenged at various time points between 12 and 96 hr under hypoxic conditions (3% 0 2 ) with and without LPS (10 pg/ml), IL-lp and tumor necrosis factor (TNF-a; 2.5 ng/ml). Expression of mRNA and protein for ecNOS, interleukin 6 (IL-6), platelet-derived growth factor (PDGF), vascular endothelial growth factor (VEGF), and endothelin-1 (ET-I) was assessed by northern and western blotting, competetive polymerase chain reaction and in situ hybridization.
