SUMMARY A method for visualizing the nucleoid in living bacteria is described. The method involves immobilization of the cells by attachment to a poly‐l‐lysine coated slide and subsequent immersion in bovine serum albumin solutions of defined refractive index and osmolality. Dependence of the visibility of the nucleoid on the refractive index of the medium is investigated.
