SUMMARY The common methods of counting Azotobacter spp. in soil, colony counts and MPN, were compared using cell suspensions of densities determined by direct microscopic counts. A highly significant positive correlation was found between the surface‐plate, MPN and total cell counts, but the MPN estimations approximated total cell counts much more closely. Inclusion of 1‐1.5 g of agar/l of the liquid medium enhanced multiplication of azotobacters and facilitated the recording of growth in MPN estimations.
