The amount of Vascular Endothelial Growth Factor (VEGF) Anti‐angiogenic isoforms to pro‐angiogenic isoforms has greater effects on follicle development and progression than pro‐angiogenic isoforms in the perinatal rat ovary

Perinatal rat ovarian organ cultures (postnatal day 3; P3) were treated with pro‐angiogenic VEGF (VEGF164) and antibodies to anti‐angiogenic isoforms (anti‐VEGFxxxb). Organ cultures treated with anti‐VEGFxxxb dramatically decreased the number of follicles that were at the primordial and early primary follicle stage (stage 0 and 1) and increased the number of follicles that were at later stages (stage 2, 3, 4) compared to controls ( P < 0.005). In contrast, treatment with the pro‐angiogenic VEGF 164 resulted in ovaries that contained less primordial follicles (stage 0) and more developing (1–4) than controls ( P < 0.04). Further experiments were conducted in vivo where P0 female rats were treated for three days with IP injections of VEGF164; VEGF165b and anti‐VEGFxxxb. There were decreased numbers of primordial follicles in ovaries from the anti‐VEGFxxxb treated rats with greater numbers of follicles that were at later stages of development (P < 0.05). Furthermore, in ovaries from rats treated with the VEGF165b there were reduced follicles that progressed to later stages of development (P<0.05). From these data we can conclude that VEGF pro and anti‐angiogenic isoforms regulate follicle development and progression. Thus, manipulation of the VEGF gene to produce different VEGF isoforms may allow for prevention of female infertility disorders such as premature ovarian failure.
