Puerarin enhanced Proliferator‐Activated Receptor γ (PPAR‐γ) activation in 3T3‐L1 preadipocytes

Of the herbal medications used in diabetic disorders, Puerariae Radix is mentioned as one of the important crude materials in oriental medicine. Diets containing the Puerariae Radix‐derived phytoestrogens, puerarin and daidzein increased plasma cholesterol and glucose levels in experimental animals. The mechanisms responsible for anti‐hyperglycemic effects of puerarin is unknown. In this study the effects of puerarin on glucose uptake and adipocyte differentiation in 3T3‐L1 cells were investigated. The effect of puerarin on intracellular glucose uptake was investigated in differentiated adipocytes using a radioactive 2‐D‐[3H]‐deoxyglucose, and the ability of puerarin to induce differentiation of preadipocytes was examined by Oil‐red‐O staining method. The effect of puerarin on expression of peroxisome proliferator‐activated receptor γ (PPAR γ) gene, which is associated with hyperglycemia, was examined by both real‐time PCR and reverse transcriptase PCR. Puerarin at the concentration of 50uM increased glucose uptake in 3T3‐L1 preadipocyte by 120%. Puerarin stimulated differentiation of 3T3‐L1 preadipocyte induced by insulin plus IBMX and DEX (IS‐IBMX‐DEX) in a dose‐dependent manner. Puerarin also resulted in significant induction of PPAR γ gene expression. Thus, puerarin has potential to be developed into an attractive adjuvant for the treatment of diabetic patients.
