The unique role of a new (SCS)Ni cofactor in lactate racemase (LarA) is examined in this computational study. The Ni‐dependent cofactor could increase dehydrogenation barriers and particularly destabilize the NADH‐like pyruvate intermediates, due to the lower electrophilicity of the cofactor and the ring strain in the intermediates. This intermediate destabilization by a cofactor in LarA, which can promote racemization, is uncommon in enzymatic catalysis. More information can be found in the Full Paper by L. W.
