O fator de crescimento vascular endotelial (VEGF: “vascular endothelial growth factor”) é um mediador chave do processo de angiogênese e está relacionado a vários processos fisiológicos e patológicos. O VEGF é uma proteína dimérica que faz parte de uma superfamília de proteínas que possuem em suas estruturas um conjunto de ligações dissulfeto denominado “nó de cistina” (cystine knot). A presença dessas ligações dissulfeto na estrutura do VEGF está relacionada à sua atividade biológica, mas não é responsável pela sua estabilidade termodinâmica. O presente estudo, dedica-se à investigação por simulações de dinâmica molecular dos efeitos de mutações de resíduos de cisteína na estrutura e na dinâmica do VEGF. Modelos modificados (i.e. mutantes) do VEGF são gerados através da troca de certos resíduos de cisteína por resíduos de alanina ou serina de forma que ligações dissulfeto selecionadas sejam rompidas. Simulações de dinâmica molecular destes modelos são conduzidas e os resultados são então comparados com aqueles da estrutura nativa (WT: wild type) e com as estruturas cristalográficas dos respectivos mutantes. As simulações indicam que a troca de resíduos de cisteína por resíduos de alanina ou serina provocam mudanças estruturais e dinâmicas localizadas, basicamente, afetam a alça-2, que está localizada na região de ligação ao receptor. DOI: 10.5935/1984-6835.20110005The vascular endothelial growth factor (VEGF) is a key mediator of angiogenesis and is related to several physiological and pathological processes. VEGF is a dimeric protein that belongs to a superfamily of proteins that including in their structures a set of disulfide bonds forming the so called cystine knot. The presence of these disulfide bonds in the structure of VEGF is related to its biological activity, but is not responsible to its thermodynamic stability. The present study aims at the investigation via molecular dynamics simulations of the effects of mutations of cysteine residues on the structure and dynamics of VEGF. Modified models (i.e. mutants) of VEGF are constructed by replacing certain cysteine residues by alanine or serine residues in such a way that selected dissulfide bonds are broken. Molecular dynamics simulations of these models are then carried out and the results are compared to the ones previously obtained for the native structure (WT: wild type) and to the respective crystalographic structures of the mutants. The simulations indicate that the replacement of cysteine residues by alanine or serine promote localized structural changes, basically affecting the loop-2, which is localized at the receptor-binding region. DOI: 10.5935/1984-6835.2011000
