Thioglycosides are widely used in the synthesis of oligosaccharides because they provide high levels of chemical stability and can be chemoselectively activated in a glycosylation reaction. Thioglycosides, however, are prepared from thiols, which can be challenging to use because of their pungent and generally unpleasant odors, as well as their tendency to form disulfides through oxidation, and these factors have effectively limited the application of thioglycosides in organic synthesis. Herein, we describe recent efforts towards the development of odorless methods for the preparation of thioglycosides, as well as an evaluation of their reactivity in the synthesis of oligosaccharides. A. Introduction Together with glycosyl halides and trichloroacetimidates, thioglycosides have become abundantly versatile glycosyl donors for the synthesis of oligosaccharides (1). The success of thioglycosides in this context has been attributed to their high level of chemical stability, with thioglycosides being compatible with most of the reaction conditions involved in the regioselective protection of the parent sugar unit. Furthermore, thioglycosides can be readily synthesized by the treatment of a thiol with a per-O-acetylated sugar in the presence of an appropriate acid or with glycosyl bromide under basic conditions. One of the most remarkable features of thioglycosides is that their thioether moiety can be orthogonally activated by a range of thiophilic chemical species such as halogenoand alkyl cations in the presence of glycosyl acceptors bearing heterogeneous aglycone, which allows for sequential glycosylation reactions. Meanwhile, as many carbohydrate chemists have testified, most thiols evolve unpleasant, pungent odors, which can make their use in the preparation thioglycosides particularly difficult.
