Orotate phosphoribosyl transferase(OPRT)酵 素 活性 とフ ッ化 ピ リ ミジ ン系抗 癌剤(5-FU)の 効 果 に は正 の相 関が あ る と報告 されて い る。今 回 われわ れは大 腸癌 に おい て,OPRT酵 素 活性 とOPRTの 翻 訳領域 に存在 す るSNPか ら,5-FU効 果 予測 の可 能性 を検 討 した。対 象 は2003年12月 ~2004年7月 の期 間 に,当 科 に て外科 的切 除 が行 われ た大腸 癌35例 。 正常 部,腫 瘍 部 のOPRT活 性値(nmol/min/mg)をradioassay法 にて 測定 し,末 梢血 の SNPはinvader法 に て解析 した。SNPは638G/C,1050T/A,1336A/Gの 三種 類 を用 い,OPRT酵 素 活性 との 関連 性 を検討 した。OPRT酵 素 活性 は正 常部 に比べ 腫 瘍部 で有意 に 高か っ た。三種 類 のSNPに お いて,OPRT酵 素 活 性 に有意 差 は認め られなか った。OPRT酵 素 活 性 と今 回の三種 類 のSNPと の 間に 関連性 は認 め られ ず,こ れ らの 今 回検討 したSNPか ら5-FU効 果 を予 測す るこ とは困難 で あ る と考 え られ た。
