生 きた肝 細 胞 の合 成 能 や 代 謝能 を積 極 的 に 利用 し肝 不 全 治 療 に 応 用 しよ う とす るバ イ オ 人工 肝 臓(bioartificial liver,以 下BALと 略 す)が 注 目され てい る。高 性能 なBALの 開発 は,創 薬や 薬物 代謝 の検 定 モデ ルに も 使 用 で きる応用 範囲 の広 い もの であ る。BALに は健常 ヒ ト細 胞が理 想 で あ るこ とは 間違 いな いが,世 界的 な ヒ ト ドナー 肝臓 の不 足か らES細 胞,肝 幹 細胞,骨 髄 細 胞,異 種 動 物 の肝臓 細胞 な どの利用 が研 究 され て いる。われ わ れ は,こ れ まで細 胞の ソー ス として取 扱 いの容 易 な ヒ ト細胞株 の樹 立 及 び当該細 胞 の分化 誘導療 法 の 開発 に努 め て きた。 本稿 で は,こ う した われ われ の一連 の研 究 と世 界 のBAL治 療 の現 状 を議論 す る。
