Typhula incarnata Lash ex Fr., one of the snow mold fungi, invades plants at a slow speed under snow. Although there are some descriptions of the penetration of this pathogen into wheat and barley1-3), the mode of penetration and the time of host penetration are still uncertain. The objectives of this study are, therefore, to determine the mode of penetration of T. incarnata, and the relationship between the penetration by this pathogen and wheat plant resistance. Part of this study has already been reported elsewhere4). The wheat cultivars used in this study were Norin No.61 and Yukichabo. Five seeds of each cultivar were planted at the opposite sides of a plastic box (13.3•~28.4•~9.0cm) containing autoclaved sandy loam soil and fertilizer (N content: 0.76g/box). The plants were grown at 10-23C in a glasshouse for 18 days and transferred to outdoors in early December to allow plants to cold-harden under natural conditions for 11 days. Inoculation was done by the methods of Bruehl et al.5) and Takamatsu6) with slight modifications. An isolate of T. incarnata (HT8301), obtained from a naturally infected barley in Niigata Prefecture, was incubated at 15C for 10 days in potato sucrose broth, transferred thereafter in sand-wheat bran-dextrose medium (sand 1,000ml, wheat bran 1,000 ml, dextrose 14g and distilled water 250ml), incubated at 15C for 17-18 days and used as an inoculum. One hundred and ten g of inoculum was distributed evenly on the soil in each plastic box. The leaves of the seedling at the 3-4 leaf stage were bent on the inoculum and covered with a mat of wet absorbent cotton. Each pot was wrapped in a polyethylene bag to keep humidity and incubated in the dark at 0-1C. After each cultivar was incubated for 4-7 weeks, the cotton was removed and the plants were replaced in the glasshouse to recover. The number of surviving plants was recorded after the 14day recovery period.
