The effects of asteltoxin, which is a toxic metabolite of Aspergillus stellatus Curzi and Emericella variecolor NHL 2881, on oxidative phosphorylation in isolated rat liver mitochondria has been examined in order to assess its cytotoxicity. It was found that this toxin specifically inhibited the energy transfer system of the oxidative phosphorylation in mitochondria. Emericella variecolor NHL2881か ら強 力 な呼 吸 毒 作 用 を 有 す る 二 次 代 謝 産 物EV-toxinが 単 離 さ れ, す で に.Aspergillus stellatus Curziか ら単 離 さ れ て い るasteltoxinと 構 造 が一 致 す る こと が判 明 した. Asteltoxinはhexatriene側 鎖 を有 す る α-pyrone誘 導 体 で,aurovertinsやcitreoviridinに 似 た構 造 を 有 し,し か も呼 吸 毒 作用 を示 す こ とか ら,こ れ ら類 似 化 合 物 と 同 様 の 生物 活 性,即 ち 単 離 肝 ミ トコ ン ド リア の 呼 吸 系 に対 す る エ ネ ル ギ ー転 移 阻 害 作用 を 示 す こ とが 推 測 され た.Asteltoxinは 大 腸 菌 のBF1-ATPase 活 性 を 強 く阻 害 す る こ とが す で に見 出 さ れ て い る が,酸 化 的 リン酸 化 反 応 の 呼 吸 鎖(電 子伝 達 反 応)へ の作 用 お よ び 除共 役 作用 の 有 無 に つ い て は未 だ 検 討 さ れ て い な い.そ こで,ラ ッ ト肝 ミ トコ ン ドリア を用 い た 実験 で,酸 化 的 リン酸 化 反 応 へ の影 響 を酸 素電 極 法 に よ り検 討 した結 果,asteltoxinが ミ トコ ン ド リア の 呼 吸 系 に対 して 特異 的 に エ ネ ル ギ ー 転 移反 応 を 阻 害 し,高 濃度 で も電 子 伝 達 系反 応 へ の 阻 害 作 用 や 除 共 役 作 用 を 示 さな い こ とが 判 明 した の で 報 告 す る.
