The in vivo and in vitro effects of vitamin E on platelet aggregation were investigated.1) Alpha-tocopherol was added to platelet rich plasma (PRP) in concentrations of 125, 250 and 500μg/ml, respectively. Dose dependent inhibitory effect of added tocopherol was observed on platelet aggregation induced by ADP and epinephrine (tocopherol concentrations in the platelets examined were 0.5-1.5μg/mg protein).2) The healthy volunteers were orally given 1200mg/day of alpha-tocopheryl nicotinate for one week. No significant inhibitory effect on platelet aggregation induced by ADP, epinephrine and collagen was observed after the tocopherol administration.3) Intramuscular administration of 600mg of alpha-tocopheryl acetate was carried out in five healthy volunteers. The platelet tocopherol elevated in the mean level of 0.45μg/mg protein. The aggregation was found to be inhibited in the platelets.The above finding suggests that intramuscular administration of tocopherol is necessary to maintain an inhibitory effect on platelet aggregation.
