Nebulin, a high molecular weight (~700 kDa) protein of vertebrate skeletal muscle, was not detected in embryonic and adult cardiac muscles of the chicken by SDS-gel electrophoresis. No peptides reactive with anti-nebulin antiserum were found by immunoblot tests. Nebulin was detected in chicken embryonic skeletal muscles after 17 days of incubation. On the other hand, connectin, another giant protein, was present in both cardiac and skeletal muscles of the chicken embryos after 15 days of incubation. Nebulin is the second largest protein (M. W., ~700 kDa) in vertebrate skeletal muscles (8), and is localized in the I band about 120 nm apart from the Z lines of chicken skeletal muscle myofrbrils (6). It has been reported that nebulin is more or less degraded in Duchenne dystrophic muscles (5, 9), although its physiological role remains unknown. We have shown that nebulin band is not detected in frog, chicken, and rabbit cardiac muscles by SDS-gel electrophoresis (2). Locker and Wild (3) also reported the absence of the nebulin by SDS-gel electrophoresis of samples from chicken, rabbit and sheep heart muscles. It is still possible that nebulin with a molecular mass other than 700 kDa exists in cardiac muscles. Another possibility is that 700 kDa nebulin is expressed in early embryonic muscles but its expression ceases in differentiated muscles (cf 4). The present investigation clearly excludes the above-mentioned possibilities using immunoblot techniques. Chicken cardiacmuscleswere gentlyhomogenized with two volumes of an SDS-solution (10% sodium dodecyl sulfate, 40 mM dithiothreitol, 10 mM EDTA and 0.1 M Tris buffer, pH 8.0) and heated for 2-3 min at 100°C.
