Geothermal sites are habitat for thermophilic microorganisms. In this study, a novel aerobic, rod shaped, motile and Gram-positive thermophilic bacterium DB2 was isolated from the hot water in Dibekli in the town of Diyadin in Turkey and it was identified by morphological, physiological, biochemical tests and 16S rRNA gene sequencing. Optimal cultivation was obtained at pH 6.0–6.5 and 60 oC. Strain DB2 was positive for catalase activity, nitrate reduction, Methyl red test and starch hydrolysis, while it was negative for oxidase, urease, gelatine and casein hydrolysis. Phylogenetic analysis based on 16S rRNA gene sequences showed that isolate DB2 was most closely related to Geobacillus subterraneus subsp. aromaticivorans Ge1(T) with a 98.97% sequence similarity. This novel strain can be used in biotechnological application.
