著者らはレーザー誘起キャピラリー振動法(CVL法)をキャピラリー電気泳動(CE)の高感度検出法として提案,開発してきた.CVL法は蛍光物質に限らず適用範囲が広い.一方,光導波エキシマレーザーなど比較的小型で使い勝手のよい紫外レーザーが近年入手可能となった.実用的な検出システムの構築を目指し,このパルス紫外レーザーをCVLの励起光源として導入した.又,メチルセルロース溶液等のポリマー溶液は,従来のゲル同様DNA断片等生体高分子の分離媒体として有効であり,ゲルと異なり流動性を持つため繰り返し使用可能という優れた特長を持つ.又,その流動性のため,尖頭値の高いパルスレーザー光照射に対し損傷を受けにくく,パルスCVLにも十分適用可能と考えた.そこでメチルセルロース溶液を分離媒体としたDNA断片のCE分離及びCVL法による高感度検出を試みた.媒体の損傷はみられず,検出下限(S/N=2)7 amolを得た.
