A simple, specific, and rapid method for the estimation of thiamine and its pyrophosphoric ester is described. Hydrolysis of the phosphate groups is not necessary. After deproteinization, the sample is passed through a column of sephadex G-25 and determined by the thiochrome test. The best reproducibility was obtained in the range 0.5—3 %. The recovery of pure substances was 97% for thiamine and 99% for cocarboxylase.
