Significance How to ensure effectors are loaded efficiently is a key question to understanding protein secretion systems. Our study demonstrates that the type VI secretion system (T6SS) ofVibrio cholerae requires the physical presence of effectors as a prerequisite for assembling a functional secretion apparatus, representing a distinct posttranslational energy-saving strategy that may be conserved among the diverse T6SS systems in gram-negative bacteria.
