Significance The activity-dependent plasticity of neurotransmission has been intensively studied for decades. Our study uses a combination of chemical analysis methods and a neuroendocrine cell line to show that plasticity can occur at the level of a single vesicle. The neurotransmitter release as well as the transmitter storage inside vesicles can be dynamically regulated, which opens the possibility for cell systems to change the fraction released from each vesicle and offers a possible link between fraction of neurotransmitter release and synaptic strength and plasticity.
