Significance Nucleosome assembly is regulated at multiple levels to impact distinct cellular processes. Defects in H3.3 incorporation into the chromatin of senescent cells lead to transcriptional reprogramming that alters gene expression. Therefore, it is important to determine how nucleosome assembly of H3.3 is controlled in senescent cells. In this paper, we provide evidence of a link between the nucleosome assembly regulator Pak2 kinase and cellular senescence and organismal aging.
