Significance Temperature dramatically affects lifespan in both vertebrates and invertebrates. Recent studies have challenged the notion that cold promotes longevity solely through thermodynamic effects. We support this idea by describing a genetic manipulation influencing the longevity response to temperature inDrosophila . Low temperatures activate 4E-BP [eukaryotic translation initiation factor 4E (eIF4E)-binding protein], triggering changes in metabolism, protein translation, and lifespan. In conjunction with recent studies, the findings support the idea that nutrition and temperature affect lifespan via partially overlapping mechanisms and highlight the 4E-BP/eIF4E pathway as a pharmacological target to treat aging-related pathologies and maximize longevity.
