Significance The spatial distributions of neuronal populations are frequently patterned, but molecular mechanisms underlying their orderliness have received little attention. We used a genetic screen to identify prospective genes participating in the patterning within a population of retinal amacrine cells. We mapped variation in their patterning, assessed using nearest-neighbor analysis across a panel of 25 recombinant inbred strains, to a genomic locus on chromosome 11. There, we identified pituitary tumor-transforming gene 1 (Pttg1 ) as a candidate. Critically,Pttg1 expression correlated with regularity across these 25 strains, and a variant inPttg1 was found to modulate expression. By knocking outPttg1 , patterning was selectively disrupted in two types of retinal neuron.
